The northbound carriageway of the A419 has been completely shut for roadworks that will last the whole weekend.
The busy dual carriageway is shut from Blunsdon to Cerney Wick and will not be reopening to motorists until the early hours of Monday morning.
Delays are expected in the area as motorists have to seek alternative routes to get to their weekend destinations.
Issues have also been caused to bus services in the area with Stagecoach West having to divert to some stops and miss others out entirely.
A spokesperson said: "Service 51, due to weekend closure of the A419, we will be diverting via Purton Stoke to Cricklade and then via Ashton Keynes to South Cerney, during this time we will be unable to service Latton, sorry for any inconvenience."
